As a completely different twist you could try the trick they used in "Ronin" (if I remember correctly).

That film revolves around a suitcase-with-contents that various terrorist/intelligence/mercenaries/other groups are after. The hero gets instructed to go after the suitcase, too, but isn't told what's in it, as it's irrelevant to his assignment. "Just get it!". As a viewer, you never get to know what it was that was in suitcase, not even at the ending.

I think it's easy to create something like a mysterious sarcophacus (does it contain religious artefacts that positively prove Jesus was the son of god (or not)? Does it contain diamonds or gold? Does it contain something magical that gives great power? A beast that can be revived in exactly 5000 years (which just happens to be tomorrow)? Modern day stuff like drugs or nukes? Any theory can come by, without you having to make a decision and tie it in to the story.

You could do something with "The spear of destiny".
The story goes that the spear used by a roman soldier to stab Jesus when he was on the cross. It apparently fell into the hands of Hitler at the beginning of WWII. When some guy called General Patton found the spear in a bunker under Nuremberg and realized what it was, Hitler, at that very moment, committed suicide. The US get the spear and then becomes the new world power.
And the spear actually exists too. It is kept in Vienna, Austria. You can find out more here: http://petragrail.tripod.com/spear.html
Wasn't the sequel to Wolfenstein 3D based off of that?
I never saw that :/ . It has been used in a couple of movies though.

An epic is a genre of film which places emphasis on human drama on a grand scale. They are more ambitious in scope than other genres which helps to differentiate them from similar genres such as the period piece or adventure film. This often entails high production values, a sweeping musical score by an acclaimed film composer, and an ensemble cast of bankable stars, placing them among the most expensive of films to produce. The genre likely derives its name from the similarities it shares with epic poetry.

You may want to PM Badger about using the ideas created for the ill-fated community project Adventure. The concept was a lot like what it sounds like you're working on. We had been working on a script for a couple of months and actually got pretty far on it, but eventually scrapped it due to lack of interest and because it wasn't particularly community friendly.

Either that or the biblical western knight monkeys thing. That sounds awesome, too.
